Elastase, Porcine pancreas弹性蛋白酶

　　弹性蛋白酶Elastase, Porcine pancreas (EC 3.4.21.36) 是具有 240 个氨基酸残基的单条多肽链，来源于猪胰腺。弹性蛋白酶Elastase, Porcine pancreas 是一种丝氨酸蛋白酶，可以水解蛋白质和多肽。Elastase from porcine pancreas 可诱发仓鼠气肿。

　　体外研究

 

　　溶液内消化方案

 

　　1. 将 Elastase 重悬于双蒸水中，最终浓度为 1 mg/mL。将重悬的 Elastase 在 4°C 下保存最多 2 周。

 

　　2. 将蛋白质重新悬浮在反应缓冲液中。

 

　　3. 将 Elastase 加入蛋白质溶液中，混合。我们建议使用 1:20 至 1:100 的酶：蛋白质比例。

 

　　4. 37°C 孵育 2-18 小时。

 

　　5. 加入10% 甲酸或 TFA 至最终浓度为 0.5% 或在 95°C 加热 10 分钟来终止反应。

 

　　体内研究

 

　　弹性蛋白酶Elastase, Porcine pancreas (EC 3.4.21.36) 会诱发仓鼠肺气肿[3]。
